(A) Curves depicting the IC50 values of [111In]In-Ex4 and its derivatives in CHL cells expressing the GLP-1R. The average IC50 values are displayed separately in the upper right-hand corner. Significant differences were calculated using the Welch ANOVA test with α < 0.05, which showed that the IC50 of all Ex4 derivatives is significantly higher than the IC50 of the reference compound [111In]In-Ex4. (B) Cell uptake and surface binding of radiolabelled Ex4 and its derivatives in CHL cells expressing the GLP-1R after 5, 15, 30, 60 and 120 min. One-way ANOVA comparing [111In]In-Ex4 derivatives to the reference compound [111In]In-Ex4 was conducted for the percentage of cell uptake at 120 min. Only [111In]In-MV-MVK-Ex4 showed significantly lower uptake than [111In]In-Ex4 after 120 min. *p < 0.05
