From: Fully automated radiosynthesis of [18F]LBT999 on TRACERlab FXFN and AllinOne modules, a PET radiopharmaceutical for imaging the dopamine transporter in human brain

1) Azeotropic drying and complex [18F]KF/K222 formation

1- [18F]Fluoride trapping on a QMA after production and transfer from the cyclotron

2- Elution of [18F]Fluoride in the reactor with a solution of K222/K2CO3

3- Azeotropic drying of [18F]Fluoride with successive addition of acetonitrile

2) [18F]radiofluorination of 4

1- Addition of the precursor 4

2- [18F]Radiofluorination

3) Purification of [18F]LBT999

1- Cooling and Dilution of the crude mixture with water or a solution of water in presence of Na-Asc (0.5%).

2- Pre-purification on C18 and washing with water or a solution of water in presence of Na-Asc (0.5%).

3- Elution of crude product and injection into the HPLC loop

4- HPLC Purification

5- Collection of the product

6- Trapping on C18 cartridge and washing

4) Formulation and dispensing of [18F]LBT999

1- Elution with 10% EtOH and saline (0.9%) or saline (0.9%) with Na-Asc (0.5%)

2- Transfer of formulated [18F]LBT999 and vial dispensing after sterile filtration
